Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_024334.3(TMEM43):c.410G>A (p.Gly137Glu), citing Ambry Variant Classification Scheme 2023: The p.G137E variant (also known as c.410G>A), located in coding exon 5 of the TMEM43 gene, results from a G to A substitution at nucleotide position 410. The glycine at codon 137 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species; however, glutamine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
