This variant lies in the TPM1 gene (transcript NM_001018005.2) at coding-DNA position 583, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 195 with lysine — a missense variant. Submitter rationale: The p.E195K variant (also known as c.583G>A), located in coding exon 6 of the TPM1 gene, results from a G to A substitution at nucleotide position 583. The glutamic acid at codon 195 is replaced by lysine, an amino acid with some similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
