Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.75271A>G (p.Ser25091Gly), citing Ambry Variant Classification Scheme 2023: The p.S16026G variant (also known as c.48076A>G), located in coding exon 153 of the TTN gene, results from an A to G substitution at nucleotide position 48076. The serine at codon 16026 is replaced by glycine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6038 samples (12076 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
