Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.50230C>G (p.Leu16744Val), citing Ambry Variant Classification Scheme 2023: The p.L7679V variant (also known as c.23035C>G), located in coding exon 93 of the TTN gene, results from a C to G substitution at nucleotide position 23035. The leucine at codon 7679 is replaced by valine, an amino acid with highly similar properties, and is located in the A-band region of the N2-B isoform of the titin protein. This variant was not reported in population-based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), Exome Aggregation Consortium (ExAC), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6082 samples (12164 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
