Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001134363.3(RBM20):c.2945G>T (p.Arg982Ile), citing Ambry Variant Classification Scheme 2023: The p.R982I variant (also known as c.2945G>T), located in coding exon 11 of the RBM20 gene, results from a G to T substitution at nucleotide position 2945. The arginine at codon 982 is replaced by isoleucine, an amino acid with some similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 2283 samples (4566 alleles) with coverage at this position. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
