Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.57519G>T (p.Lys19173Asn), citing Ambry Variant Classification Scheme 2023: The p.K10108N variant (also known as c.30324G>T), located in coding exon 121 of the TTN gene, results from a G to T substitution at nucleotide position 30324. The lysine at codon 10108 is replaced by asparagine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
