Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001035.3(RYR2):c.11217G>A (p.Met3739Ile), citing Ambry Variant Classification Scheme 2023: The p.M3739I variant (also known as c.11217G>A), located in coding exon 81 of the RYR2 gene, results from a G to A substitution at nucleotide position 11217. The methionine at codon 3739 is replaced by isoleucine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6135 samples (12270 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
