NM_003476.5(CSRP3):c.233G>T (p.Gly78Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the CSRP3 gene (transcript NM_003476.5) at coding-DNA position 233, where G is replaced by T; at the protein level this means replaces glycine at residue 78 with valine — a missense variant. Submitter rationale: The p.G78V variant (also known as c.233G>T), located in coding exon 2 of the CSRP3 gene, results from a G to T substitution at nucleotide position 233. The glycine at codon 78 is replaced by valine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6492 samples (12984 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
