t(6;17)(p21.1;q24.3) was classified as Likely pathogenic for Skeletal dysplasia; Sex reversal; Kyphoscoliosis; Hydrocephalus; Respiratory distress; Camptomelic dysplasia by Kahle Lab, Yale University, citing Antwi et al. (Cold Spring Harb Mol Case Stud. 2018): The translocation is upstream of SOX9, the only gene known to have pathogenic variants in campomelic dysplasia. This translocation, like those previously reported, results in under-expression of SOX9, leading to the disease phenotype.
