This variant lies in the PKP2 gene (transcript NM_001005242.3) at coding-DNA position 2213, where T is replaced by C; at the protein level this means replaces valine at residue 738 with alanine — a missense variant. Submitter rationale: The p.V782A variant (also known as c.2345T>C), located in coding exon 12 of the PKP2 gene, results from a T to C substitution at nucleotide position 2345. The valine at codon 782 is replaced by alanine, an amino acid with similar properties. This variant was detected in a cardiomyopathy/arrhythmia genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected in some cases (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
