NM_001134363.3(RBM20):c.2761A>T (p.Ile921Phe) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I921F variant (also known as c.2761A>T), located in coding exon 11 of the RBM20 gene, results from an A to T substitution at nucleotide position 2761. The isoleucine at codon 921 is replaced by phenylalanine, an amino acid with highly similar properties. This variant was reported in individual(s) with features consistent with dilated cardiomyopathy (van der Meulen MH et al. Circ Genom Precis Med, 2022 Oct;15:e002981). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
