Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.0001 in 251310 control chromosomes, predominantly at a frequency of 0.0014 within the East Asian subpopulation in the gnomAD database. This frequency is not significantly higher than estimated for a pathogenic variant in COL11A2 causing COL11A2-Related Disorders, allowing no conclusion about variant significance. To our knowledge, no occurrence of c.2529+13delG in individuals affected with COL11A2-Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported.
