Uncertain significance — the classification assigned by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine to NM_000256.3(MYBPC3):c.926+1G>A, citing LMM Criteria. This variant lies in the MYBPC3 gene (transcript NM_000256.3) at the canonical splice donor site of the intron immediately after coding-DNA position 926, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant classified as Uncertain Significance - Favor Pathogenic. The c.926+1G>A variant in MYBPC3 has not been previously reported in individuals with cardiomyopathy but has been identified in 3/27668 of South Asian chromosomes by gnomAD (http://gnomad.broadinstitute.org). This variant occurs in the invariant region (+/- 1,2) of the splice consensus sequence and is predicted to cause altered splicing leading to an abnormal or absent protein. In summary, while there is some suspicion for a pathogenic role, the clinical significance of this variant is uncertain. ACMG/AMP Criteria applied: PVS1_Moderate, PP3.
