NM_000059.4(BRCA2):c.1825C>T (p.Gln609Ter) was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q609* pathogenic mutation (also known as c.1825C>T), located in coding exon 9 of the BRCA2 gene, results from a C to T substitution at nucleotide position 1825. This changes the amino acid from a glutamine to a stop codon within coding exon 9. This alteration has been reported with a carrier frequency of 0.00013 in 7636 unselected prostate cancer patients and 0.00 in 12366 male controls of Japanese ancestry (Momozawa Y et al. J Natl Cancer Inst, 2020 04;112:369-376). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
