Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.93G>T (p.Gln31His), citing Ambry Variant Classification Scheme 2023: The p.Q31H variant (also known as c.93G>T), located in coding exon 1 of the FBN2 gene, results from a G to T substitution at nucleotide position 93. The glutamine at codon 31 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
