Uncertain significance — the classification assigned by Genetic Services Laboratory, University of Chicago to NM_006231.4(POLE):c.2908G>A (p.Gly970Ser), citing ACMG Guidelines, 2015: DNA sequence analysis of the POLE gene demonstrated a sequence change, c.2908G>A, in exon 25 that results in an amino acid change, p.Gly970Ser. This sequence change does not appear to have been previously described in patients with POLE-related disorders. This sequence change is absent from the large population databases (ExAC and gnomAD). The p.Gly970Ser change affects a highly conserved amino acid residue located in a domain of the POLE protein that is known to be functional. The p.Gly970Ser substitution appears to be deleterious using several in-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL). Due to these contrasting evidences and the lack of functional studies, the clinical significance of the p.Gly970Ser change remains unknown at this time.
