NM_033629.6(TREX1):c.748dup (p.Thr250fs) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.748dupA variant in the TREX1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.748dupA variant causes a frameshift starting with codon Threonine 250, changes this amino acid to an Asparagine residue, and creates a premature Stop codon at position 12 of the new reading frame, denoted p.Thr250AsnfsX12. This variant is predicted to cause loss of normal protein function through protein truncation, as the last 65 amino acids are lost and replaced with 11 incorrect amino acids. The c.748dupA variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.748dupA as a likely pathogenic variant.
