NM_001042492.3(NF1):c.7869+5G>C was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the NF1 gene (transcript NM_001042492.3) at 5 bases into the intron immediately after coding-DNA position 7869, where G is replaced by C. Submitter rationale: The c.7806+5G>C variant in the NF1 gene has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016). Several in silico splice prediction models predict that c.7806+5G>C destroys the natural donor site of intron 52 and lead to abnormal gene splicing. However, in the absence of RNA/functional studies, the actual effect of this sequence change in this individual is unknown. Based on the currently available information, it is unclear whether this variant is a pathogenic variant or a rare benign variant. We consider it to be a variant of uncertain significance.
