NM_000368.5(TSC1):c.1030-14T>A was classified as Uncertain significance for Tuberous sclerosis syndrome by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This variant causes a T to A nucleotide substitution at the -14 position of intron 10 of the TSC1 gene. Splice site prediction tools suggest that this variant may have a significant impact on RNA splicing, although this prediction has not been confirmed in published RNA studies. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with TSC1-related disorders in the literature. This variant has been identified in 3/282376 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
