Likely pathogenic — the classification assigned by GeneDx to NM_004281.4(BAG3):c.258C>G (p.Tyr86Ter), citing GeneDx Variant Classification (06012015): The Y86X variant in the BAG3 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Y86X variant is not observed in large population cohorts (Lek et al., 2016). We interpret Y86X as a likely pathogenic variant.
