NM_005219.5(DIAPH1):c.2905_2906dup (p.Ser970fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the DIAPH1 gene (transcript NM_005219.5) at coding-DNA position 2905 through coding-DNA position 2906, duplicating 2 bases; at the protein level this means shifts the reading frame starting at serine residue 970, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.2905_2906dupAA pathogenic variant in the DIAPH1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The c.2905_2906dupAA variant causes a frameshift starting with codon Serine 970, changes this amino acid to a Arginine residue, and creates a premature Stop codon at position 9 of the new reading frame, denoted p.Ser970ArgfsX9. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.2905_2906dupAA variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.2905_2906dupAA as a pathogenic variant.
