Pathogenic — the classification assigned by GeneDx to NM_032638.5(GATA2):c.345dup (p.Trp116fs), citing GeneDx Variant Classification (06012015): The c.345dupC variant in the GATA2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant causes a frameshift starting with codon Tryptophan 116, changes this amino acid to a Leucine residue, and creates a premature Stop codon at position 69 of the new reading frame, denoted p.Trp116LeufsX69. The c.345dupC variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. This variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.345dupC as a pathogenic variant.
