It causes a frameshift starting with codon Serine 41, changes this amino acid to an Arginine residue and creates a premature Stop codon at position 63 of the new reading frame, denoted p.Ser41ArgfsX63. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). In summary, we consider this variant to be pathogenic.
