Pathogenic — the classification assigned by GeneDx to NM_005422.4(TECTA):c.1131_1134del (p.Ser376_Trp377insTer), citing GeneDx Variant Classification (06012015). This variant lies in the TECTA gene (transcript NM_005422.4) at coding-DNA position 1131 through coding-DNA position 1134, deleting 4 bases. Submitter rationale: The c.1131_1134delGGTG variant in the TECTA gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.1131_1134delGGTG variant causes a frameshift at codon Tryptophan 377, changing this amino acid to a premature Stop codon, denoted p.Trp377Ter. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.1131_1134delGGTG variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.1131_1134delGGTG as a pathogenic variant.
