Likely pathogenic — the classification assigned by GeneDx to NM_001854.4(COL11A1):c.2485_2486insAAG (p.Ser828_Gly829insGlu), citing GeneDx Variant Classification (06012015). This variant lies in the COL11A1 gene (transcript NM_001854.4) at coding-DNA position 2485 through coding-DNA position 2486, inserting AAG. Submitter rationale: The c.2485_2486insAAG variant in the COL11A1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant causes an in-frame insertion of a Glutamic acid residue, denoted p.Ser282_Gly829insGlu. The c.2485_2486insAAG variant is not observed in large population cohorts (Lek et al., 2016). In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret c.2485_2486insAAG as a likely pathogenic variant.
