This variant lies in the GNAI1 gene (transcript NM_002069.6) at coding-DNA position 832, duplicating one base; at the protein level this means shifts the reading frame starting at isoleucine residue 278, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.832dupA variant has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.832dupA variant causes a frameshift starting with codon Isoleucine 278, changes this amino acid to a Asparagine residue, and creates a premature Stop codon at position 20 of the new reading frame, denoted p.Ile278AsnfsX20. This variant is predicted to cause loss of normal protein function through protein truncation as the last 77 amino acids are lost and replaced with 19 incorrect amino acids. The c.832dupA variant is not observed in large population cohorts (Lek et al., 2016).
