This variant lies in the FH gene (transcript NM_000143.4) at coding-DNA position 221, duplicating one base; at the protein level this means shifts the reading frame starting at serine residue 75, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This duplication of one nucleotide in FH is denoted c.221dupG at the cDNA level and p.Ser75IlefsX6 (S75IfsX6) at the protein level. The normal sequence, with the base that is duplicated in brackets, is GTGA[dupG]ATCT. The duplication causes a frameshift which changes a Serine to an Isoleucine at codon 75, and creates a premature stop codon at position 6 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. We consider this variant to be pathogenic.
