Pathogenic — the classification assigned by GeneDx to NM_000088.4(COL1A1):c.2484_2510dup (p.Glu828_Gly836dup), citing GeneDx Variant Classification (06012015). This variant lies in the COL1A1 gene (transcript NM_000088.4) at coding-DNA position 2484 through coding-DNA position 2510, duplicating 27 bases. Submitter rationale: The c.2484_2510dup27 pathogenic variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge, but has been observed de novo in an affected individual tested at GeneDx. The c.2484_2510dup27 variant results in the in-frame duplication of 9 amino acids starting with codon Glutamic acid 828, denoted p. Glu828_Gly836dup. The duplicated amino acids are within the Gly-X-Y repeat within the triple helical region of the protein. Variants affecting these repeats result in poor winding of the collagen triple helix and a less functional protein. Multiple in-frame variants adding or removing Gly-X-Y repeats have been reported in the Human Gene Mutation Database in association with COL1A1-related disorders (Stenson et al., 2014).
