Pathogenic — the classification assigned by GeneDx to NM_000047.3(ARSL):c.123TCT[1] (p.Leu43del), citing GeneDx Variant Classification (06012015): The c.126_128delTCT variant in the ARSE gene has been reported previously as hemizygous in a male with brachytelephalangic chondrodysplasia punctata or X-linked chondrodysplasia punctata 1 (CDPX1) (Matos-Miranda et al., 2013). The c.126_128delTCT variant is not observed in large population cohorts (Lek et al., 2016). In vitro expression assays demonstrated that c.126_128delTCT retained only residual activity of ARSE, compared to wild type (Matos-Miranda et al., 2016). We interpret c.126_128delTCT as a pathogenic variant.
