Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 108992 control chromosomes. c.1063G>T has been observed in individuals affected with Osteogenesis Imperfecta (e.g. Fahiminiya_2013, Bardai_2016). At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect resulted in <10% of normal ability to stimulate WNT signalling (Palomo_2014).
