NM_001267550.2(TTN):c.48727C>G (p.Pro16243Ala) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P7178A variant (also known as c.21532C>G), located in coding exon 87 of the TTN gene, results from a C to G substitution at nucleotide position 21532. The proline at codon 7178 is replaced by alanine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and alanine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
