Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.4417G>A (p.Glu1473Lys), citing Ambry Variant Classification Scheme 2023: The p.E1473K variant (also known as c.4417G>A), located in coding exon 27 of the CFTR gene, results from a G to A substitution at nucleotide position 4417. The glutamic acid at codon 1473 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
