This variant lies in the PEX16 gene (transcript NM_004813.4) at coding-DNA position 461, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 154 with glycine — a missense variant. Submitter rationale: The PEX16 c.461A>G variant is predicted to result in the amino acid substitution p.Asp154Gly. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.084% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/11-45936235-T-C), which may be too common to be an undocumented primary cause of disease. Although we suspect that this variant may be benign, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
