Uncertain significance for Muscular dystrophy-dystroglycanopathy (congenital with brain and eye anomalies), type A9; Autosomal recessive limb-girdle muscular dystrophy type 2P — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004393.6(DAG1):c.1454G>A (p.Ser485Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with asparagine at codon 485 of the DAG1 protein (p.Ser485Asn). The serine residue is moderately conserved and there is a small physicochemical difference between serine and asparagine. This variant is present in population databases (rs764859263, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with DAG1-related conditions. ClinVar contains an entry for this variant (Variation ID: 500607). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
