Four of four in-silico tools predict a damaging effect of the variant on protein function. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.1566T>G has been reported in the literature in a homozygous individual affected with Niemann-Pick Disease (Panigrahi_2019) and has been observed in unknown individual(s) with this disease (Almeida_2022). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
