Pathogenic for Sitosterolemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022436.3(ABCG5):c.1256G>A (p.Arg419His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 419 of the ABCG5 protein (p.Arg419His). This variant is present in population databases (rs119479067, gnomAD 0.009%). This missense change has been observed in individual(s) with sitosterolemia (PMID: 11452359, 11855938, 21576934). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 4978). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. For these reasons, this variant has been classified as Pathogenic.
