Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_002474.3(MYH11):c.1301G>A (p.Arg434His), citing Ambry Variant Classification Scheme 2023: The p.R434H variant (also known as c.1301G>A), located in coding exon 11 of the MYH11 gene, results from a G to A substitution at nucleotide position 1301. The arginine at codon 434 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
