Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000344.4(SMN1):c.835-29T>G, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the SMN1 gene (transcript NM_000344.4) at 29 bases into the intron immediately before coding-DNA position 835, where T is replaced by G. Submitter rationale: Variant summary: SMN1 c.835-29T>G is located at a position not widely known to affect splicing. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 242914 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.835-29T>G in individuals affected with Spinal Muscular Atrophy and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as VUS-possibly benign.
