Uncertain significance for Primary dilated cardiomyopathy — the classification assigned by Agnes Ginges Centre for Molecular Cardiology, Centenary Institute to NM_000719.7(CACNA1C):c.1582T>G (p.Trp528Gly), citing ACMG Guidelines, 2015: The CACNA1C Trp528Gly is a novel rare variant being absent from both population databases: 1000 genomes project (http://www.1000genomes.org/) and Exome Aggregation Consortium dataset (http://exac.broadinstitute.org/). In silico tools predict that the protein to be pathogenic (SIFT, PolyPhen-2, MutationTaster). We identified this variant in a male proband presenting with sudden unexplained death at 32yo. Due to the lack of information we have classified CACNA1C Trp528Gly as a variant of 'uncertain significance'.
