Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000455.5(STK11):c.1063G>C (p.Asp355His), citing Ambry Variant Classification Scheme 2023: The p.D355H variant (also known as c.1063G>C), located in coding exon 8 of the STK11 gene, results from a G to C substitution at nucleotide position 1063. The aspartic acid at codon 355 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
