This variant lies in the SMAD4 gene (transcript NM_005359.6) at 6 bases into the intron immediately after coding-DNA position 424, where T is replaced by C. Submitter rationale: This variant causes a T to C nucleotide substitution at the +6 position of intron 3 of the SMAD4 gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 3/251232 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
