This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 531, where A is replaced by T; at the protein level this means replaces glutamic acid at residue 177 with aspartic acid — a missense variant. Submitter rationale: The p.E177D variant (also known as c.531A>T), located in coding exon 3 of the MSH2 gene, results from an A to T substitution at nucleotide position 531. The glutamic acid at codon 177 is replaced by aspartic acid, an amino acid with highly similar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was reported to be functionally neutral (Jia X et al.
