Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000251.3(MSH2):c.530A>G (p.Glu177Gly), citing Ambry Variant Classification Scheme 2023: The p.E177G variant (also known as c.530A>G), located in coding exon 3 of the MSH2 gene, results from an A to G substitution at nucleotide position 530. The glutamic acid at codon 177 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. In addition, this alteration is predicted to be deleterious by MAPP-MMR in silico analyses (Chao EC et al. Hum.
