NM_007194.4(CHEK2):c.710C>A (p.Ala237Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A237D variant (also known as c.710C>A), located in coding exon 5 of the CHEK2 gene, results from a C to A substitution at nucleotide position 710. The alanine at codon 237 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
