Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000077.5(CDKN2A):c.313G>A (p.Asp105Asn), citing Ambry Variant Classification Scheme 2023: The p.D105N variant (also known as c.313G>A), located in coding exon 2 of the CDKN2A gene, results from a G to A substitution at nucleotide position 313. The aspartic acid at codon 105 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
