This variant lies in the BRIP1 gene (transcript NM_032043.3) at coding-DNA position 837, where G is replaced by T; at the protein level this means replaces arginine at residue 279 with serine — a missense variant. Submitter rationale: This sequence change replaces arginine with serine at codon 279 of the BRIP1 protein (p.Arg279Ser). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and serine. This variant is present in population databases (rs759584091, ExAC 0.001%). This variant has not been reported in the literature in individuals with BRIP1-related disease.
