Likely pathogenic — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_032043.3(BRIP1):c.1888dup (p.Thr630fs), citing Quest Diagnostics criteria: This frameshift variant is predicted to cause the premature termination of BRIP1 protein synthesis. It has been reported in a couple of individuals with breast cancer as well as healthy controls in the published literature PMIDs: 26921362 (2016) and 33471991 (2021)). The frequency of this variant in the general population is consistent with pathogenicity. Based on the available information, we predict that the variant is likely pathogenic.
