NM_000465.4(BARD1):c.1917T>G (p.Cys639Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C639W variant (also known as c.1917T>G), located in coding exon 10 of the BARD1 gene, results from a T to G substitution at nucleotide position 1917. The cysteine at codon 639 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
