Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.5250G>C (p.Trp1750Cys), citing Ambry Variant Classification Scheme 2023: The p.W1750C variant (also known as c.5250G>C), located in coding exon 34 of the ATM gene, results from a G to C substitution at nucleotide position 5250. The tryptophan at codon 1750 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
