Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024675.4(PALB2):c.676A>G (p.Thr226Ala), citing Ambry Variant Classification Scheme 2023: The p.T226A variant (also known as c.676A>G), located in coding exon 4 of the PALB2 gene, results from an A to G substitution at nucleotide position 676. The threonine at codon 226 is replaced by alanine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
